BioNTech's BNT113 Misses Progression-Free Survival Endpoint, but the Trial Isn't Over Yet

When a biotech company stakes its future on a therapeutic area as unforgiving as oncology, the market's first reaction to disappointing data tends to be swift and blunt. The nuance arrives later — if it arrives at all.
That pattern played out again this week for BioNTech, the Mainz-based company trying to reinvent itself as a broad cancer immunotherapy player after its pandemic-era windfall. Shares fell 7.6% on Tuesday, closing at EUR 79.40 and widening the gap to the 52-week high of EUR 105.80 to 25%. The trigger was preliminary data from the Phase II/III AHEAD-MERIT trial, which is testing the mRNA candidate BNT113 in combination with the antibody pembrolizumab as a first-line treatment for advanced, HPV16-positive head and neck cancer.
At the first analysis point, the combination failed to demonstrate a statistically significant extension of progression-free survival compared with pembrolizumab alone — one of two co-primary endpoints. The miss stung, particularly coming on the heels of the company's decision in late August to terminate a Phase II study of the candidate autogene cevumeran as a monotherapy in patients with operated colorectal cancer.
A Setback, Not a Verdict
Yet missing that first statistical hurdle does not spell the end of the program. In oncology, the initial endpoint is rarely the final word on a drug candidate. The independent data monitoring committee reviewed the interim results and explicitly recommended continuing the trial. Attention now shifts to the second dual primary endpoint: overall survival.
Should investors sell immediately? Or is it worth buying BioNTech?
Under the pre-specified study design, a statistically significant survival benefit alone would be sufficient to formally declare the trial a success. BioNTech remains blinded to the data, so details such as the hazard ratio, median progression-free survival, and the exact timing of the final analysis are being kept under wraps for now.
The company's leadership has been here before. CEO Ugur Sahin reported sales of 36,209 and 27,000 ordinary shares, transactions carried out under a pre-arranged trading plan adopted on June 3, 2026, in accordance with Rule 10b5-1. Such disclosures tend to draw attention regardless of their mechanical nature, and this one was no exception — a reminder that formal automatisms rarely neutralize emotional responses during sensitive moments.
Analysts Keep Their Composure
Institutional observers have responded with more equanimity than the trading floor. Evercore ISI analyst Cory Kasimov maintained his "Outperform" rating and a price target of USD 130 per share. The progression-free survival miss was not entirely unexpected given earlier study data, he noted, pointing to more encouraging trends in overall survival and stressing that the stock's trajectory depends heavily on other advanced pipeline projects such as Pumitamig.
Citi also left its buy recommendation and USD 125 price target unchanged, arguing that convincing overall survival data could still secure the development path for BNT113.
BioNTech holds an advantage few competitors in the biotech sector can match: time. As of June 30, 2026, cash, cash equivalents, and securities investments totaled approximately EUR 16.63 billion. With 14 oncology studies running as of the summer, the pipeline is broad enough to absorb setbacks on individual candidates.
At a share price of EUR 79.95, the company carries a market capitalization of EUR 20.99 billion. The transformation into a diversified oncology operation is proving a demanding journey, one on which interim steps must be assessed soberly. In biotechnology, the only verdict that ultimately counts is robust clinical proof of efficacy — and for BNT113, that verdict is still pending.
