BioNTech's BNT113 Setback Leaves Overall Survival as the Deciding Card

BioNTech's attempt to reinvent itself as a broad-based cancer immunotherapist is proving to be a far tougher slog than its sprint to a pandemic-era vaccine. Tumor biology does not bend to deep pockets or lofty expectations, and the latest readout from the Mainz-based biotech's pipeline has driven that lesson home once again.
Shares in the German company came under heavy selling pressure on Tuesday, closing down 7.6% at EUR 79.40. That widens the gap to the 52-week high of EUR 105.80 to 25%. The trigger was preliminary data from the Phase II/III AHEAD-MERIT trial, which is testing the mRNA candidate BNT113 alongside the antibody pembrolizumab as a first-line treatment for advanced, HPV16-positive head and neck cancer.
At the first analysis point, the combination failed to demonstrate a statistically significant extension of progression-free survival compared with pembrolizumab alone — one of two co-primary endpoints missed. The disappointment landed hard on investors, particularly since the company had only wrapped up a Phase II study of the candidate Autogene Cevumeran as a monotherapy in operated colorectal cancer patients at the end of August.
The Trial Isn't Over Yet
Does the miss on the first goal spell the end of the program? Far from it. After reviewing the interim results, the independent data monitoring committee explicitly recommended that the study continue. Attention now shifts to the second dual primary endpoint: overall survival.
Under the pre-specified trial design, a statistically significant survival benefit on its own would be enough to formally crown the study a success. BioNTech remains blinded to the ongoing data, so specifics such as the hazard ratio, median progression-free survival, and the exact timing of the final analysis are being kept under wraps for now.
Should investors sell immediately? Or is it worth buying BioNTech?
The decision point on the drug therefore rolls forward to the next evaluation. That nuance has drawn a more measured response from institutional observers than from the trading floor. Evercore ISI analyst Cory Kasimov reaffirmed his "Outperform" rating and a USD 130 price target, noting that the progression-free survival miss was not entirely unexpected given earlier study data. He also pointed to more encouraging overall survival trends and stressed that the stock's path from here hinges largely on other advanced pipeline projects such as Pumitamig.
Citi was equally unmoved, keeping its buy recommendation and a USD 125 target, arguing that convincing overall survival data could still keep the development path for BNT113 intact.
Insider Sales Follow a Pre-Set Script
Running alongside the clinical news, BioNTech disclosed securities transactions by its leadership. CEO Ugur Sahin sold shares in several tranches in early October, including 17,000 shares at USD 97.4556 apiece on October 1, with further disposals in the days that followed.
All of the transactions were executed under a Rule 10b5-1 trading plan that Sahin put in place on June 3, 2026. Such arrangements are designed to lock in executive sales well in advance and on fixed terms, with the trades processed automatically regardless of current clinical events or short-term market swings.
Time Is the One Asset Competitors Can't Match
BioNTech holds an advantage few peers in biotech can claim: time. As of June 30, 2026, its cash, cash equivalents, and securities investments stood at roughly EUR 16.63 billion. With 14 oncology studies running as of the summer, the pipeline is broad enough to absorb setbacks on individual candidates.
Even so, the latest data make plain that the road to market approval in oncology remains strewn with statistical hurdles and scientific unknowns. For BioNTech shareholders, the long haul that translational medicine demands is simply part of the bargain — and the final verdict on BNT113 has yet to be delivered.
